Yes, aperture then ISO, unless you are concerned with DoF. Then ISO first. We can discuss the effect of aperture on picture sharpness in the intermediate thread.

Basically. You need a heavy piece of paper, something that won't allow the light to shine through. It also needs to fill the frame, but focus isn't an issue.
